Detect leucine aminopeptidase activity in streptococci and related genera with Thermo Scientific™ Remel™ LAP Disk.
Collman and Ball investigated the use of leucine aminopeptidase (LAP) that helped to identify Gram-positive cocci1. Further, Fertally and Facklam2 used LAP to identify Streptococcus spp. They found that Aerococcus is a consistent negative strain for LAP whereas Enterococcus and Streptococcus are positive strains. LAP also helps in identification of Leuconostoc, Pediococcus, Aerococcus and related genera3.
Activity of the enzyme leucine aminopeptidase can help in identifying streptococci and related genera using the LAP Disk.

- Easy to read: Visible color change indicates positive and negative test.
Positive test: Color change from pink to red.
Negative test: No color change.
The LAP Disk contains L-leucine-β-naphthylamide as substrate that is hydrolyzed by leucine aminopeptidase and releases β-naphthylamide. A positive test is indicated when the color of the disk changes from white to pink to red when ρ-dimethyl-aminocinnamaldehyde (PYR Reagent) is added.
