Preparation: Produced from sera of rabbits immunized with highly pure Recombinant Human/Murine/Rat BDNF. Anti-Human/Murine/Rat BDNF-specific antibody was purified by affinity chromatography and then biotinylated. Sandwich ELISA: To detect Human BDNF by sandwich ELISA (using 100 μL/well antibody solution) a concentration of 0.25-1.0 μg/mL of this antibody is required. This biotinylated polyclonal antibody, in conjunction with PeproTech Polyclonal Anti-Human BDNF (500-P84) as a capture antibody, allows the detection of at least 0.2-0.4 ng/well of Recombinant Human BDNF. Western Blot: To detect hBDNF by Western Blot analysis this antibody can be used at a concentration of 0.1-0.2 μg/mL. Used in conjunction with compatible secondary reagents the detection limit for Recombinant hBDNF is 1.5-3.0 ng/lane, under either reducing or non-reducing conditions. 500-P84BT-1MG will be provided as 2 x 500 μg
BDNF (Brain-Derived Neurotrophic Factor) is a member of the neurotrophin family. BDNF is synthesized as pre-proBDNF, followed by cleavage to proBDNF. Although further processing generates the mature, 14 kDa protein, pro-BDNF is biologically active and is secreted in synaptic vesicles along with the mature form. BDNF is widely expressed in the central nervous system and acts in an autocrine and paracrine manner on several classes of neurons. Signaling occurs mainly through the tyrosine kinase receptor TrkB, although binding to the lower-affinity receptor, p75-NTR, has also been demonstrated. BDNF promotes neuronal survival and differentiation and has been shown to play a critical role in memory formation and synaptic regulation. BDNF is induced by cortical neurons, and is necessary for survival of striatal neurons in the brain. Expression of BDNF is reduced in both Alzheimer's and Huntington disease patients, and may also play a role in the regulation of stress response and in the biology of mood disorders.Specifications
